What can you do with 1000 base reads?
- Continuous development of the robust 454 Sequencing platform will enable the Genome Sequencer FLX Titanium chemistry to make the next leap in read length, breaking the 500 base pair mark and approaching 1000 bases.
- Even longer single reads will sequence through more repeat regions, co-locate more exons and link more associated variants, opening the full spectrum of structural variation analysis and significantly improve de novo sequencing, transcriptomics, metagenomics and resequencing applications.
- Combine super long single reads with our unique 20kb, 8kb, 3kb paired end reads to generate high-quality draft assemblies with more complete genome coverage.

The first steps towards simplifying the 454 Sequencing workflow began with the recent introduction of the GS FLX Titanium Series reagents. Concepts such as large volume emulsions, reduced packaging, fewer component tubes and simplified protocol volumes, plus a completely reworked method for emulsion breaking using a semi-automated vacuum breaking device and centrifugation.
- Automation of the emPCR enrichment and sequencing primer annealing steps will simplify the next most time consuming and labor intensive part of emPCR.
- The compact enrichment device will integrate with most common bench top liquid handling robots and perform the critical functions of washing, vortexing, heating, cooling and magnetic bead separation.
- Free up users for other important tasks with complete walk-away automation for 2-region LV and 4-region SV emulsions, while 8- and 16-region emulsions will require only one quick intervention to change filters.
- Cut sample preparation time significantly. Finish enrichment and annealing in about ½ the time. Start sequencing today instead of tomorrow.
