- How much input DNA is required?
- 5ug of double stranded DNA.
- What quality of DNA is required?
- Provide the DNA at a concentration > 300ng/ul and A260/280 ratio of approximately 1.8. Providing an image of DNA sample(s) run on a 2% agarose gel is requested to assess quality of incoming DNA.
- What coverage is needed?
- De novo sequencing requires 20X average genome coverage. Resequencing and mapping project require 15X average coverage.
- How long do projects take?
- Length of a project depends on number of samples, genome size and data analysis requirements. Typical bacterial projects are normally completed within a month of receipt of sample.
- Has anyone done a project like mine before?
- Browse through a selected list of projects completed at 454 in the Services section. Fill out a Request for Service form to talk to one of our account managers for more detail.
- How should samples be shipped?
- Click here for sample submission guidelines (.pdf)
- What should the DNA be resuspended in?
- H2O or 1XTE is preferred.
- How much input DNA is required? And volume?
- 5ug of double stranded DNA, no less than 10 ul.
- What If my DNA is 500 bp long or less, how much do you really need?
- At that point there is no nebulization. 2 ug is enough, no less than 10 ul in volume.
